Nutritional factors influencing the polysaccharide production of Brucella suis.

چکیده
The formation of cellular polysaccharide by cells of the genus Brucella has been reported by several investigators: Favilli and Biancalani (1934), Higginbotham and Heathman (1936), and Libby and Joyner (1941). These workers, however, dealt mainly with the immunological characteristics of the polysaccharide, and little emphasis was placed upon the nutritional conditions affecting its production. A comparison of the differences in the media with the chemical characteristics of the polysaccharide material isolated from Brucella abortus, Brucella suis, and Brucella melitensis by Huston et al. (1934) and Higginbotham and Heathman (1936) suggests that nutrition may play an important role in the synthesis of polysaccharide by this group of organisms. Investigations in this laboratory with B. suis revealed certain nutritional conditions which significantly altered the polysaccharide content of this organism. The data suggest that this polysaccharide was intracellular and therefore probably unrelated to the immunologically active fractions studied by others. The following presentation is concerned primarily with identification of the nutritional conditions affecting polysaccharide formation. The isolation and chemical characterization of the polysaccharide will be reported later.

The Epitopic and Structural Characterization of Brucella suis Biovar 2 O-Polysaccharide Demonstrates the Existence of a New M-Negative C-Negative Smooth Brucella Serovar
The brucellae are Gram-negative bacteria that cause an important zoonosis. Studies with the main Brucella species have shown that the O-antigens of the Brucella smooth lipopolysaccharide are α-(1 → 2) and α-(1 → 3)-linked N-formyl-perosamine polysaccharides that carry M, A and C (A = M, A>M and A<M) epitopes relevant in serodiagnosis and typing. We report that, in contrast to the B. suis biovar...
متن کاملThermostable cross-protective subunit vaccine against Brucella species.
A subunit vaccine candidate was produced from Brucella suis 145 (biovar 4; expressing both the A antigen of Brucella abortus and the M antigen of Brucella melitensis). The preparation consisted mostly of polysaccharide (PS; >90% [wt/wt]; both cell-associated PS and exo-PS were combined) and a small amount of protein (1 to 3%) with no apparent nucleic acids. Vaccinated mice were protected (these...
متن کاملExpression and bactericidal activity of nitric oxide synthase in Brucella suis-infected murine macrophages.
We examined the expression and activity of inducible nitric oxide synthase (iNOS) in both gamma interferon (IFN-gamma)-treated and untreated murine macrophages infected with the gram-negative bacterium Brucella suis. The bacteria were opsonized with a mouse serum containing specific antibrucella antibodies (ops-Brucella) or with a control nonimmune serum (c-Brucella). The involvement of the pro...
متن کاملProduction and Characterization of Monoclonal Antibodies against Brucella abortus S (99) Surface Antigens
By immunizing mice with killed whole bacterial cells of Brucella abortus S (99), a panel of six hybridomas producing monoclonal antibodies (mAb) specific for the surface antigens of this bacterium were produced. ELISA was used to screen the hybridoma supernatants. Immunoblots of the cell extract indicated that three mAb were specific for S-LPS (Ba-1, Ba-2, Ba-3) and three others were reactive w...
متن کاملFactors influencing utilization of the stereoisomers of homocystine by Brucella.
Previous studies (Lankford et al., 1952) demonstrated that minute quantities of Dor i-methionine (0.001 to 0.01 ug S per ml) exert a stimulatory effect on the utilization of "DL" (inactive)-homocystine by selected strains of Brucella suis. The growth response to D-homocystine was found to be proportional to the quantity of methionine present, either as contamination of the commercial sample, or...
